Regulatory treatment varies by country. In the United States, melanotan-2 is not approved for any indication, and products marketed for human use fall outside the approved drug framework. Some other jurisdictions have placed it under prescription controls or listed it as a prohibited or restricted substance. Online listings frequently describe the material as a research chemical, a category that does not carry the same manufacturing and labelling requirements as approved medicines.
Solid peptide material is generally stable when kept cold and dry. Common practice is storage at -20 degrees Celsius or lower, with desiccant and protection from light. Repeated freeze-thaw cycles and exposure to moisture are associated with degradation, aggregation, or loss of material. Once dissolved, stability depends on solvent, concentration, and temperature, and solutions are usually treated as short-lived unless stability data support longer periods. Handling notes typically emphasise minimising time at ambient temperature.
Identity and purity are assessed with chromatographic and mass spectrometric techniques. Reversed-phase high-performance liquid chromatography separates the target peptide from related impurities and degradation products, and the resulting retention time is compared against a reference standard. Mass spectrometry, often coupled to liquid chromatography, confirms molecular mass. Amino acid analysis or peptide mapping can provide additional sequence-level confirmation when required. Results are only as reliable as the reference materials used alongside them.
Published pharmacokinetic information is limited and comes mainly from small studies rather than registrational trials. Plasma half-life is usually described as short, on the order of tens of minutes, followed by rapid tissue distribution and clearance of the intact peptide. Metabolites and low concentrations of parent compound have been reported in urine, a detail relevant to anti-doping and forensic testing. Whether repeated exposure changes receptor sensitivity or clearance over time remains an open question. Values differ noticeably between analytical assays, so published numbers should be read as approximate rather than definitive.
Melanotan II is a synthetic cyclic heptapeptide with the sequence Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, corresponding to a molecular formula of C50H69N15O9 and a monoisotopic mass near 1024 daltons. It was designed as a structural analogue of alpha-melanocyte-stimulating hormone, a peptide hormone produced by cleavage of proopiomelanocortin. A lactam bridge between the aspartate and lysine side chains closes the ring, and the C-terminal amide removes a free carboxyl group. Both modifications increase resistance to enzymatic degradation compared with the linear parent hormone. Four substitutions distinguish it from afamelanotide, the linear analogue studied under the name melanotan I.
Receptor-binding studies classify melanotan II as a non-selective melanocortin agonist. It interacts with MC1R, MC3R, MC4R and MC5R, with reported affinities in the low nanomolar range and no strong subtype preference. Activation of MC1R on dermal melanocytes shifts pigment synthesis toward eumelanin, the dark polymer deposited in melanosomes and transferred to keratinocytes. Because the same peptide engages MC4R in the hypothalamus, it also appears in animal work on food intake and erectile response, which is why it is discussed in both pigment and metabolic research. Which receptor populations dominate after systemic exposure in humans is not fully established.

Because the substance circulates mainly through informal markets, verification is a recurring theme in technical discussion. Independent analyses have found that labeled content and actual content can diverge, and that purity varies between samples. Analytical laboratories use reversed-phase chromatography to separate components and mass spectrometry to confirm identity. Isotope-labeled internal standards improve quantification in complex matrices. Such methods describe what a sample contains but say nothing about its sterility, lawful status, or suitability for any use. Open questions remain about how consistently testing is applied across the supply chain.
Regulatory treatment of this peptide varies by country. It holds no marketing authorization as a medicine in the United States, the European Union, or most other jurisdictions. Some countries classify products containing it as prescription-only or unlicensed medicines, which restricts lawful supply. Authorities have issued public notices warning that unregulated products may contain undeclared or incorrect ingredients. The molecule also appears on prohibited lists for competitive sport. These measures address supply oversight rather than any approved therapeutic role.
Published research on the compound remains limited. Much of the human data comes from small, early-stage studies rather than large controlled trials, and several questions about effects and variability between individuals remain open. Investigators have examined receptor activity, pigment pathways, and related physiological responses in laboratory and animal models. Findings from those models do not automatically translate to human outcomes. Reviews frequently note the scarcity of rigorous clinical evidence and call for better-characterized study material.
Melanotan II binds several melanocortin receptor subtypes rather than a single target. MC1R on melanocytes drives melanin synthesis, while MC3R and MC4R participate in energy balance, appetite, and sexual response pathways. This lack of selectivity explains why reported effects extend beyond skin darkening. Substitutions at positions four and seven, including norleucine and D-phenylalanine, increase potency and resistance to peptidases. Understanding which receptor mediates which effect remains an active area of investigation.
Published human data come mostly from small, short studies rather than large controlled trials. Reported outcomes include increased skin pigmentation and, in some reports, effects on appetite and libido, but sample sizes are small and follow-up is limited. Whether long-term use produces durable pigment changes or adverse effects is not established. Because products sold outside pharmacies are not standardized, the actual content of any given vial is often unknown. Independent testing of such material is uncommon.
Melanotan II is a synthetic cyclic heptapeptide analog derived from the core sequence of alpha-melanocyte-stimulating hormone. Researchers at the University of Arizona synthesized it during the 1980s while studying pigmentation and appetite signaling. The compound is not an approved medicine in any major jurisdiction and appears mainly in laboratory and research-chemical settings. Its structure incorporates a lactam bridge between side chains, which constrains the ring and slows enzymatic breakdown relative to the natural hormone.
Regulatory treatment varies by country. In the United States the peptide is not approved as a medicine, and products offered for human use may be treated as unapproved new drugs; some states also restrict sale. Australia, the United Kingdom, and European Union member states apply comparable restrictions to unapproved peptide products. Border agencies have seized shipments labelled as research chemicals. Classification may change over time, and the legal position for personal importation is not clearly settled in most published guidance.
Lyophilised melanotan-2 is supplied as a solid, which is more stable than a solution. The material is hygroscopic, so weighing is done quickly, in low humidity, with the container kept sealed. Reconstitution usually uses water for injection or bacteriostatic water, added down the wall of the vial to limit foaming. A reconstituted solution is held at 2 to 8 °C and kept away from light. Repeated freezing and thawing of the same vial is avoided because ice crystal formation and concentration effects degrade the peptide.
Pharmacologically, melanotan-2 behaves as a non-selective agonist across the melanocortin receptor family. Binding at MC1R on dermal melanocytes promotes eumelanin synthesis, which underlies the tanning response described in early human work. Activity at the centrally expressed MC4R receptor is associated with reported effects on appetite and erectile function. Because the peptide does not discriminate strongly among receptor subtypes, attributing any single observed effect to one receptor pathway is generally not possible without selective antagonists or receptor knockout models.
Research interest has centred on photoprotection and pigmentation, with a smaller body of work on appetite and sexual function. Published human data remain limited to small, frequently uncontrolled studies, and the compound has never received marketing approval from a national medicines regulator. Most laboratory work treats it as a pharmacological tool for probing melanocortin signalling in cell culture or animal models. Whether pigmentation changes observed in people translate into measurable protection against ultraviolet-induced DNA damage remains an open question.
In a sense, the study of biochemistry can be considered to have started in ancient times, for example when biology first began to interest society—as the ancient Chinese developed a system of medicine based on yin and yang, and also the five phases, which both resulted from alchemical and biological interests. Its beginning in the ancient Indian culture was linked to an interest in medicine, as they developed the concept of three humors that were similar to the Greeks' four humours (see humorism). They also delved into the interest of bodies being composed of tissues. The ancient Greeks' conception of biochemistry was linked with their ideas on matter and disease, where good health was thought to come from a balance of the four elements and four humors in the human body. As in the majority of early sciences, the Islamic world contributed significantly to early biological advancements as well as alchemical advancements; especially with the introduction of clinical trials and clinical pharmacology presented in Avicenna's The Canon of Medicine. On the side of chemistry, early advancements were heavily attributed to exploration of alchemical interests but also included: metallurgy, the scientific method, and early theories of atomism. In more recent times, the study of chemistry was marked by milestones such as the development of Mendeleev's periodic table, Dalton's atomic model, and the conservation of mass theory. This last mention has the most importance of the three due to the fact that this law intertwines chemistry with thermodynamics in an intercalated manner.
=== Role in cancer progression and therapy resistance === ITGA1 contributes to tumor progression by regulating survival, migration, invasion, and resistance to anticancer therapies. In Pancreatic Ductal Adenocarcinoma (PDAC), ITGA1-mediated collagen EMT signaling promotes the survival of mesenchymal tumor cell populations, metastatic dissemination, and resistance to gemcitabine treatment. Inhibition of ITGA1 increases sensitivity to chemotherapy, making treatment more effective and supporting its capacity as a potential therapeutic target. In glioma, ITGA1 contributes to temozolomide (TMZ) resistance by activating the PI3K/AKT/Bcl-2 signaling pathway. Increased ITGA1 activity promotes anti-apoptotic signaling which prevents tumor cells from apoptosizing therefore leading to survival of tumor cells. Suppressing ITGA1 increases apoptosis and improves TMZ sensitivity. ITGA1 expression in glioma is regulated by the hsa-miR01298-5p pathway, which directly binds the ITGA1 3' untranslated region to suppress ITGA1 expression. Increased ITGA1 reverses those effects, making resistance to chemotherapy more likely.
==== Type 1 ==== Type 1 VWD (40-80% of all VWD cases) is a quantitative defect which is heterozygous for the defective gene. It arises from failure to secrete VWF into the circulation or, in the case of Type 1C, from VWF being cleared more quickly than normal. If VWF levels are greater than 50%, VWD can be ruled out. If VWF activity is below 30%, VWD is present. Patients with VWF activity between 0.30-0.50 IU/mL are classified based on their bleeding phenotype. If they have bleeding symptoms, they have VWD. If they have no bleeding, they are considered to have "Low VWF". Many patients are asymptomatic or may have mild symptoms and not have clearly impaired clotting, which might suggest a bleeding disorder. Often, the discovery of low VWF occurs incidentally to other medical procedures requiring a blood work-up. Most cases of low VWF are never diagnosed due to its asymptomatic or mild presentation, and most people with type I VWD end up leading a normal life free of complications, with many being unaware that they have the disorder. Trouble may, however, arise in some patients in the form of bleeding following surgery (including dental procedures), noticeable easy bruising, or menorrhagia (heavy menstrual periods). The minority of cases of type 1 may present with severe hemorrhagic symptoms.
== History == Tanning hide into leather involves a process which permanently alters the protein structure of skin, making it more durable and less susceptible to decomposition and coloring. The place where hides are processed is known as a tannery.
=== Confirmed discovery === The now-confirmed discovery of flerovium was made in June 1999 when the Dubna team repeated the first reaction from 1998. This time, two atoms of flerovium were produced; they alpha decayed with half-life 2.6 s, different from the 1998 result. This activity was initially assigned to 288Fl in error, due to the confusion regarding the previous observations that were assumed to come from 289Fl. Further work in December 2002 finally allowed a positive reassignment of the June 1999 atoms to 289Fl. In May 2009, the Joint Working Party (JWP) of IUPAC published a report on the discovery of copernicium in which they acknowledged discovery of the isotope 283Cn. This implied the discovery of flerovium, from the acknowledgement of the data for the synthesis of 287Fl and 291Lv, which decay to 283Cn. The discovery of flerovium-286 and -287 was confirmed in January 2009 at Berkeley. This was followed by confirmation of flerovium-288 and -289 in July 2009 at Gesellschaft für Schwerionenforschung (GSI) in Germany. In 2011, IUPAC evaluated the Dubna team's 1999–2007 experiments. They found the early data inconclusive, but accepted the results of 2004–2007 as flerovium, and the element was officially recognized as having been discovered.

3,14-Diacetyloxymorphone and its relatives including acetylmorphone do not, however, have annual production quotas published by the DEA in the Federal Register.[Citation Needed] Like all or most of the direct morphine derivatives, halogenated derivatives of these drugs and their hydromorphone and hydromorphinol analogues were synthesized in the 1930s when both the esters and the halogenated morphine derivatives were being developed, including one given as 1,2-iodo-3,6,14-triacetyl-6ɑ-14β-hydroxydihydromorphinone in a footnote to a 1948 German medical journal article about the esters of morphine. It appears that this drug was used, labelled with Iodine 129, as a tracer in animal studies, was significantly stronger than morphine, and possibly has 1- and/or 2- fluoro, chloro, and bromo analogues.[Citation Needed] 3,6-Diacetyloxymorphone is a third acetylated oxymorphone derivative, the oxymorphone analogue of acetylmorphone and expected to be intermediate in strength betwixt the two aforementioned drugs. Another is 3-acetyloxymorphone.
=== Military === Amid the blockade, American reconnaissance flights near Cuba increased. On 17 May, Axios reported classified U.S. intelligence that Cuba had acquired over 300 attack drones from Russia and Iran since 2023 and "recently began discussing plans" to attack the U.S. with them. Cuba described the case as "fraudulent" and a pretext for U.S. military action.

=== Air purifier distribution === On Friday, July 21, 2023, Illinois Governor JB Pritzker and the Illinois Department of Public Health announced they were partnering with SHIELD Illinois to distribute air purifiers to licensed Illinois Day Care outside of the city of Chicago. The purifiers were purchased with $10 million of federal funds through the CDC's Epidemiology and Laboratory Capacity for Prevention and Control (ELC) Reopening Schools Program. SHIELD Illinois will use the extensive logistics capabilities developed serving over 2,300 locations during the pandemic to distribute the filters across the state.
The N-hydroxyphthalimide can be removed by shaking with sodium bicarbonate, but the N-hydroxysuccinimide approach shows greater reactivity and convenience, and is generally preferred. Esters of N-hydroxyphthalimide and activated sulfonic acids such as trifluoromethanesulfonic anhydride or p-toluenesulfonyl chloride are used as so-called photoacids, which split off protons during UV irradiation.
The performance of these batteries depends on the characteristics of the material that makes up the cathode. Carbon materials have been widely used as cathodes because of their excellent electrical conductivities, large surface areas, and chemical stability. Especially relevant for lithium-air batteries, carbon materials act as substrates for supporting metal oxides. Binder-free electrospun carbon nanofibers are particularly good potential candidates to be used in electrodes in lithium-oxygen batteries because they have no binders, have open macroporous structures, have carbons that support and catalyze the oxygen reduction reactions, and have versatility. Zhu et al. developed a novel cathode that can store lithium and oxygen in the electrode they named nanolithia which is a matrix of carbon nanofibers periodically embedded with cobalt oxide. These cobalt oxides provide stability to the normally unstable superoxide-containing nanolithia. In this design, oxygen is stored as LiO2 and does not convert between gaseous and solid forms during charging and discharging. When the battery is discharging, lithium ions in nanolithia and react with superoxide oxygen the matrix to form Li2O2, and Li2O. The oxygen remains in its solid state as it transitions among these forms. The chemical reactions of these transitions provide electrical energy. During charging, the transitions occur in reverse.
Spongin, a modified type of collagen protein, forms the fibrous skeleton of most organisms among the phylum Porifera, the sponges. It is secreted by sponge cells known as spongocytes. Spongin gives a sponge its flexibility. True spongin is found only in members of the class Demospongiae. Its molecular structure remains incompletely characterized, however it shares similarities with both collagen and keratin.

Laboratory confirmation typically combines retention time matching on a chromatographic system with mass measurement. A reference standard of known identity is needed for a meaningful comparison. Sequence-level techniques can add further confirmation.
There is no single harmonised standard governing these products, and manufacturing conditions differ. Impurities may include truncated sequences, oxidation products, residual solvents, and counter-ions. Independent testing is often the only way to characterise a given lot.
Reported studies have looked at pigmentation, photoprotection, and metabolic or appetite-related endpoints in small experimental settings. Much of the evidence base is early-stage and limited in size. Questions about long-term effects and clinical relevance remain open.
No. It is a synthetic analogue carrying four amino acid changes, a lactam ring and an amidated C-terminus. The natural hormone is a linear thirteen-amino-acid peptide processed from proopiomelanocortin.
