== Calculations using calibration curve == Many analysts do not employ analytical equations for isotope dilution analysis. Instead, they rely on building a calibration curve from mixtures of the natural primary standard (A*) and the isotopically enriched standard (the spike, B). Calibration curves are obtained by plotting measured isotope ratios in the prepared blends against the known ratio of the sample mass to the mass of the spike solution in each blend. Isotope dilution calibration plots sometimes show nonlinear relationships and in practice polynomial fitting is often performed to empirically describe such curves. When calibration plots are markedly nonlinear, one can bypass the empirical polynomial fitting and employ the ratio of two linear functions (known as Padé approximant) which is shown to describe the curvature of isotope dilution curves exactly.
== Effect of plasma protein binding == For substances that exhibit substantial plasma protein binding, clearance is generally dependent on the total concentration (free + protein-bound) and not the free concentration. Most plasma substances have primarily their free concentrations regulated, which thus remains the same, so extensive protein binding increases total plasma concentration (free + protein-bound). This decreases clearance compared to what would have been the case if the substance did not bind to protein. However, the mass removal rate is the same, because it depends only on concentration of free substance, and is independent on plasma protein binding, even with the fact that plasma proteins increase in concentration in the distal renal glomerulus as plasma is filtered into Bowman's capsule, because the relative increases in concentrations of substance-protein and non-occupied protein are equal and therefore give no net binding or dissociation of substances from plasma proteins, thus giving a constant plasma concentration of free substance throughout the glomerulus, which also would have been the case without any plasma protein binding. In other sites than the kidneys, however, where clearance is made by membrane transport proteins rather than filtration, extensive plasma protein binding may increase clearance by keeping concentration of free substance fairly constant throughout the capillary bed, inhibiting a decrease in clearance caused by decreased concentration of free substance through the capillary.
Enobosarm, also formerly known as ostarine and by the developmental code names GTx-024, MK-2866, and S-22, is a selective androgen receptor modulator (SARM) which is under development for the treatment of androgen receptor-positive breast cancer in women and for improvement of body composition (e.g., prevention of muscle loss) in people taking GLP-1 receptor agonists like semaglutide. It was also under development for a variety of other indications, including treatment of cachexia, Duchenne muscular dystrophy, muscle atrophy or sarcopenia, and stress urinary incontinence, but development for all other uses has been discontinued. Enobosarm was evaluated for the treatment of muscle wasting related to cancer in late-stage clinical trials, and the drug improved lean body mass in these trials, but it was not effective in improving muscle strength. As a result, enobosarm was not approved and development for this use was terminated. Enobosarm is taken by mouth. Known possible side effects of enobosarm include headache, fatigue, anemia, nausea, diarrhea, back pain, adverse lipid changes like decreased high-density lipoprotein (HDL) cholesterol levels, changes in sex hormone concentrations like decreased testosterone levels, elevated liver enzymes, and liver toxicity, among others. The potential masculinizing effects of enobosarm, for instance in women, have largely not been evaluated and are unknown. == Directed self-assembly == Directed self-assembly (DSA) is a type of directed assembly which utilizes block co-polymer morphology to create lines, space and hole patterns, facilitating for a more accurate control of the feature shapes. Then it uses surface interactions as well as polymer thermodynamics to finalize the formation of the final pattern shapes. To control the surface interactions enabling sub-10 nm resolution, a team consisting of Massachusetts Institute of Technology, University of Chicago, and Argonne National Laboratory developed a way to use vapor-phase deposited polymeric top layer on the block co-polymer film in 2017. The DSA is not a standalone process, but rather is integrated with traditional manufacturing processes in order to mass-produce micro and nano structures at a lower cost. Directed self-assembly is mostly used in the semiconductor and hard drive industries. The semiconductor industry uses this assembly method in order to be able increase the resolution (trying to fit in more gates), while the hard drive industry uses DSA to manufacture "bit patterned media" according to the specified storage densities.
