Libya had long supported the FROLINAT militia in neighbouring Chad, but FROLINAT became divided over its ties to Libya in 1976. In January 1978, the anti-Libya faction within FROLINAT, led by Hissène Habré, switched sides and allied with Chadian President Félix Malloum. Meanwhile, the pro-Libya faction within FROLINAT, led by Goukouni Oueddei, renamed itself People's Armed Forces (FAP). In December 1980, Gaddafi reinvaded Chad at the request of the FAP-controlled GUNT government to aid in the civil war; in January 1981, Gaddafi suggested a political merger. The Organisation of African Unity (OAU) rejected this and called for a Libyan withdrawal, which came in November 1981. The civil war resumed, and Libya sent troops back in. In 1982, the GUNT government was overthrown by Habré's forces and Oueddei fled to Libya, where Gaddafi provided him with arms to continue to guerrilla war against Habré. In November 1984, Gaddafi met with French President François Mitterrand; both agreed to withdraw from Chad. Oueddei broke with Gaddafi in 1985 due to the former's intentions to negotiate a truce with Habré. Consequently, he was placed under house arrest by Gaddafi and allegedly arrested by Libyan police and shot in the stomach. Oueddei survived the shooting and fled to Algeria, but continued to claim he and Gaddafi enjoyed a good relationship. === Escherichia coli === A popular system utilized is Escherichia coli because of its rapid growth rate (~20–30 minutes), capacity for continuous fermentation and relatively low cost. Additionally, yeast has the capacity to express a high relative volume of heterologous protein. Specifically, up to 30% of proteins produced in yeast can be the heterologous gene product. There also are safe strains of E. coli that have been successfully generated to scale up production. In addition to E. coli's attractive host properties, this host is incredibly popular due to researchers having a large amount of knowledge about its genetics, including the complete genomic sequence. === Laboratory synthesis === The chemical synthesis of DMT has been described. It can be synthesized through several possible pathways from different starting materials. The two most commonly encountered synthetic routes are through the reaction of indole with oxalyl chloride followed by reaction with dimethylamine and reduction of the carbonyl functionalities with lithium aluminium hydride to form DMT. The second commonly encountered route is through the N,N-dimethylation of tryptamine using formaldehyde followed by reduction with sodium cyanoborohydride or sodium triacetoxyborohydride. Sodium borohydride can be used but requires a larger excess of reagents and lower temperatures due to it having a higher selectivity for carbonyl groups as opposed to imines. Procedures using sodium cyanoborohydride and sodium triacetoxyborohydride (presumably created in situ from cyanoborohydride though this may not be the case due to the presence of water or methanol) also result in the creation of cyanated tryptamine and beta-carboline byproducts of unknown toxicity while using sodium borohydride in absence of acid does not. Bufotenine, a plant extract, can also be synthesized into DMT. Alternatively, an excess of methyl iodide or methyl p-toluenesulfonate and sodium carbonate can be used to over-methylate tryptamine, resulting in the creation of a quaternary ammonium salt, which is then dequaternized (demethylated) in ethanolamine to yield DMT. Some longitudinal incisions of 2mm of depth have to be made through the skin to proceed to the extraction of latex, which can be collected in solid form some minutes after the extraction. The proteases present in the latex of the fruit are inactive precursors that are activated once the papaya is wounded. In 0.3 ml of latex there are about 15 mg of chymopapain. If we want to conserve the proteolytic properties, latex has to be preserved with sodium metabisulfite and stored at a low temperature of about -10 °C. If used immediately after the incisions, a buffer is added to extract the proteins: EDTA, ammonium sulfate or phosphate buffer all with a concentration of 0.5 mM and a pH of 7. It is also important to block the thiol functions to avoid air oxidation and the loss of proteolytic activity. To eliminate organic and insoluble molecules, the sample is first filtered and afterwards centrifuged at 11000g for 30min. The pellet is discarded and the supernatant added to 96% alcohol with a ratio of 1:3. Impurities precipitate and can be eliminated by filtration. Afterwards, (NH4)2SO4 fractioning is done by addition of this substance at a concentration of 0.472 mg/ml. Chymopapain precipitates and can be retrieved through another centrifugation, again at 11000g for 30min. The supernatant is discarded and the ion exchange chromatography can be carried out, with a linear gradient of 100mM (Na+) and different volumes of elution. == Murders and assaults == While she was a nurse at Caressant Care, Wettlaufer began injecting some of the patients she cared for with insulin. In some cases, the amount was not enough to kill the patient; she was charged with, and confessed to, aggravated assault or attempted murder for those cases. Wettlaufer's first assaults occurred sometime between June 25 and December 31, 2007. She confessed that she injected sisters Clotilde Adriano (age 87) and Albina Demedeiros (88) with insulin. While they later died, their deaths were not attributed to Wettlaufer. She confessed to two counts of aggravated assault. The first case in which Wettlaufer injected a patient with enough insulin to directly cause death was on August 11, 2007, when she murdered James Silcox (84), a World War II veteran and father of six.
